LA 27 begins at a junction with LA 14 in Holmwood, Louisiana, in Calcasieu Parish heading due south and at a point begins running alongside "Indian Bayou Canal". LA 27 intersects with LA 397, and 11 miles (17.7 km) south of Holmwood a traveler can visit the 9621 acres (38.9 km²) Cameron Prairie National Wildlife Refuge
