FYVE domain
Encyclopedia
FYVE zinc finger domain is named after the four cysteine-rich protein
Protein
Proteins are biochemical compounds consisting of one or more polypeptides typically folded into a globular or fibrous form, facilitating a biological function. A polypeptide is a single linear polymer chain of amino acids bonded together by peptide bonds between the carboxyl and amino groups of...

s: Fab 1 (yeast orthologue of PIKfyve), YOTB, Vac 1 (vesicle transport protein), and EEA1
EEA1
EEA1 is Early Endosome Antigen 1 protein that binds phospholipid vesicles containing phosphatidylinositol 3-phosphate, which is necessary for endosomal trafficking. It binds lipids through its FYVE domain and forms a homodimer through a coiled coil domain....

, in which it has been found.

The FYVE finger binds two zinc
Zinc
Zinc , or spelter , is a metallic chemical element; it has the symbol Zn and atomic number 30. It is the first element in group 12 of the periodic table. Zinc is, in some respects, chemically similar to magnesium, because its ion is of similar size and its only common oxidation state is +2...

ions. The FYVE finger has eight potential zinc coordinating cysteine positions and is characterized by having basic amino acids around the cysteines. Many members of this family also include two histidines in a motif R+HHC+XCG, where + represents a charged residue and X any residue.

FYVE domains bind Phosphatidylinositol 3-phosphate
Phosphatidylinositol 3-phosphate
Phosphatidylinositol 3-phosphate is a phospholipid found in cell membranes that helps to recruit a range of proteins, many of which are involved in protein trafficking, to the membranes...

, in a way dependent on its metal ion coordination and basic amino acids. It has been speculated these bindings cause conformational changes or regulate protein-protein or lipid-protein interactions.

The FYVE finger is structurally similar to the RING domain and the PHD finger
PHD finger
The PHD finger was discovered in 1993 as a Cys4-His-Cys3 motif in the homeodomain protein HAT3 in Arabidopsis thaliana. The PHD finger motif resembles the metal binding RING domain and FYVE domain...

.

The FYVE domain has been connected to vacuolar protein sorting and endosome function.
