Educated at Cambridge University, Barker first practised law in Braddell Brothers and later in Lee & Lee. He was persuaded to enter politics by Lee Kuan Yew

 and in 1963, he was made Speaker of Singapore's Legislative Assembly.

Barker's real involvement with politics also began in that same year, about a week before the elections. After Barker's victory (and later successive victories), Barker remained as Member of Parliament

, a post that he held until 1988. In his 25 years in parliament, Barker also served as the Minister for National Development (1965-75), Minister for Home Affairs (1972), Minister for Environment (1975-79), Science and Technology (1977-81) and Minister for Labour (1983).

Barker was the son of Clarence Barker and Dorothy Evaline Paterson. He was married to Gloria Hyacinth Quintal and had four children.
