U.S. Highway 27 Alternate is a signed north-south but primarily east-west route running between Perry and Williston, and is co-signed with US 19/98 between Perry and Chiefland. As part of the US 19/98/ALT 27 concurrency, it is mostly signed secretly as State Road 55. East of Chiefland, the road secretly runs along the west end of State Road 500. The entire length of the route is .
It is one of the primary routes between northwest and central Florida, having been recently been four-laned along its entire route.

It is one of the primary routes between northwest and central Florida, having been recently been four-laned along its entire route. With a signed speed limit of , this gives it more capacity than the mostly-two-lane "regular" US 27.

Former Alternate US 27
Three other Alternate U.S. Route 27's also used to exist in Florida. One around Lake Weir, from Belleview to Lady Lake was deleted in 1980. This was co-signed with Alternate U.S. Route 441 and hidden Florida State Road 25. Another in Polk County from Haines City to Sunray Deli Estates which was deleted in 1998, despite the fact that road maps and atlases still continue to show it. And a third from Avon Park to Sebring Southgate that was deleted in 1979. Both of them were replaced by formerly hidden Florida State Road 17.
