The reservation is the smallest in New York State. It is located in Mastic on the north side of Poospatuck Creek, on the east side of Poospatuck Lane, and south of Eleanor Avenue. Poospatuck is situated in the southeast corner of Suffolk County's Town of Brookhaven; and is the township's sole Indian reservation.

There are 93 households out of which 47.3% have children under the age of 18 living with them, 29.0% are married couples living together, 32.3% have a woman whose husband does not live with her, and 26.9% are non-families. 24.7% of all households are made up of individuals and 2.2% have someone living alone who is 65 years of age or older. The average household size is 2.91 and the average family size is 3.51.

In the Indian reservation the population is spread out with 36.5% under the age of 18, 10.0% from 18 to 24, 30.3% from 25 to 44, 17.3% from 45 to 64, and 5.9% who are 65 years of age or older.
