 and Route 404 (Begin Expressway) to central Jerusalem. At its junction with Route 404, the officially numbered "45" takes a 90-degree turn eastwards running 0.8 km to the Atarot Industrial Zone.

Originally, the road was supposed originate in the Dan Region, passing slightly north of Beit Horon

. The current Route 443, just south of these towns, has replaced that plan. Furthermore, an easterly extension is still officially envisioned which would continue from Atarot and connect with the existing Route 437 leading to Mishor Adummim.
