I have just started with SPR to detect interaction between two proteins. I am using GLM sensor chips for the immobilization of one of my protein (ligand protein)and flowing another analyte protein (10 amino acids long peptide) through the flow channel. I am getting good immobilization of my ligand protein as detected by resonance units but resonance units obtained during analyte injection were not good enough to account for interaction. Although, I have already checked through in vitro experiments that these two proteins interact with each other. What should I do next to optimize the interaction between my two proteins using SPR?

Hi Bhavna,
My name is venu. I am an Indian guy who is doing his PHD in Biochemistry. I just started working with SPR and it s going terribly wrong. Even my ligand immobilization seems to be non specific. There is no body in my institute who works with this machine and nobody has an idea how the data should look like. I am looking for some help and i came across your post in this forum.
