I have cells seeded over a 3D hydrogel, my aim is to visualize the cell adhesion and differentiation on the scaffold. But I fail many attempts.

I have tried fixation with gluteraldehyde, paraformaldehyde and formaldehyde. I am using anti-vinculin, rhodamine-phalloidin and hoeschst for flurosence.
